Ultra-Low Temperature Freezer Storage: Best Practices for −80 °C Samples

Best practices for −80 °C ultra-low temperature freezer storage — temperature mapping, alarms, backup power, loading, defrosting and sample organisation for labs in India.

Set-up

  • Leave at least 15 cm of clearance at the back and sides. Ambient heat is the main reason ULT compressors fail early in Indian summers.
  • Use a dedicated power circuit with a UPS or generator changeover, and an independent temperature alarm with remote (SMS or app) notification.
  • Map temperature at installation, empty and loaded, for GMP/GLP use.

Daily use

  • Use inventory racks and a sample map so the door is open for seconds, not minutes.
  • Don't overload with warm samples. Pre-cool large loads at −20 °C.
  • Clear frost from door gaskets weekly, and defrost fully as frost builds up.

Choosing a freezer

ESCO Lexicon® II ULT freezers use hydrocarbon refrigerants for efficiency, with multiple inner doors to limit warm-air ingress.

Frequently asked questions

Should a ULT freezer be set to −80 °C or −70 °C?

Many biobanks now use −70 °C for most samples. It significantly reduces energy use and compressor wear with no measurable impact on sample integrity for most material. Follow your validated storage requirement.
